Because of the team size the Sinister Six could easily be their own team.

The only question is who would be on Spidey team opposing them, here's my team:

Spidey
Venom
Peni Parker
Punisher (throwing him in because he first appeared in ASM)
Black Cat (not yet in the game but hopefully she will be someday)
Silver Sable (same situation as Black Cat, hopefully we will get her eventually)
